The stereochemistry of molecules has its roots in the work of the nineteenth‐century French physicist Jean Biot. Biot was studying the nature of plane‐polarized light (Figure 1 ) when he discovered that solutions of some organic compounds caused polarized light to rotate. What is a dextrorotatory compound? Optical activity is measured by a polarimeter, and is dependent on several factors: concentration of the sample, temperature, length of the sample tube or cell, and wavelength of the light passing through the sample. Rotation is given in +/- degrees, depending on whether the sample has d- (positive) or l- (negative) enantiomers. You can only find optical activity from experimentation. However, you will notice that if you know the optical activity for one structure, the enantiomer of that compound will have the opposite optical activity.
